For some unknown reason, my trunk will not open. The remote won’t open it. The trunk button won’t open, and the button on driver door won’t activate it. Any help would be much appreciated.

Scan for codes, see if the J393 is reporting any electrical issue with the latch motor.
You can lock out the rear lid release button in the menu interface, but I believe the fob should still open it.
I saw the comment about valet switch, but I don't believe US models have the valet switch. Maybe it got added in later years (mine being an 09).

Along with what SMAC said I'm sure you know that you can fold down the rear seats and crawl into the back and use the emergency release to open the trunk. Once you get it open have somebody operate the trunk release in the door and listen next to the latch and see if you hear anything? Before that I would do as SMAC said and do a scan. If neither the key nor the trunk release in the door work there's a good chance it's just the actuator at the trunk. If you can hear the actuator and it doesn't open it could be just the linkage. There's a little plastic tab in there that looks to be fragile. I do have a '13 S5 & RS5, same mechanism as yours and they have been both reliable.
