Here are the part numbers for the donut spare for the RS7P w/DRC & Ceramic Brakes.
The Donut will only fit on the rear, without spacer, due to 420mm caliper width on the front.
So, if you do have a front flat, no spacer, you will have to move a rear tire/wheel to the front.

Wheel spacer is needed on front, to mount donut, on RS7P with Ceramic 420 rotor for adequate caliper clearance. 5mm-8mm.

Foam Donut and Tool holder: 4G0-012-109-F, or -E for Air Suspension + wiring cover.
Wheel: 4G0-601-027-E.
Tire: DT0-350-634. Continental CST 145/60/R20 26.7 OD.
CST 165/60/R20 27.8 OD. Tight fit into compartment

Pump and Sealant fit underneath.

50 mph max.

Same wheel.
S6 shows to have a 400mm rotor?
Are your rotors OEM?

So your car is a US S6, with EU RS6 brakes? Curious about the wheel diameter and offset?

Yes. US S6 running EU RS6 brakes. On my 3rd set of custom 3 piece wheels, all have been 21x10.5" square (front and rear) (believe the ET is 34). Factory 19" wheel wouldn't clear the EU RS6 brakes, a few different 20" factory Audi wheels have cleared. (SQ5 and I think the other factory 20" were off of 2014 Allroad).Always assumed the stock donut spare wouldn't clear the EU RS6 brakes. Recently blew a front tire, jacked the car up by the front jack point high enough to remove front and rear wheel so I could swap the rear to the front and put the donut spare on the back and on a whim decided to see if the spare would fit up front and it did. Drove the car 5 miles home with no issue.

Donut without spacer will not adequately clear RS7P front calipers.
